    As a post-doctoral research associate I was involved in a fascinating project aiming to decipher the function of ISE2, a chloroplast localized RNA helicase. My interests went far beyond ISE2 and chloroplasts as obtained results demonstrate these organelles have a profound contribution to plasmodesmata biogenesis, thus also sink-to-source transition and carbohydrates allocation. For a longer run this exciting project creates a basis for new strategies in plants engineering to help cope with increasing food demands!

    • The project required me to use on a daily basis molecular biology, biochemistry, microbiology and cell biology techniques e.g.:
      - isolation of plasmid DNA,
      - DNA agarose gel electrophoresis,
      - isolation of high quality plant genomic DNA (for Southern blotting and PCR)
      - isolation of high quality RNA (for Northern blotting and RT-PCR)
      - Qubit fluorometric quantification of nucleic acids,
      - DNA cloning and restriction analysis, 
      - PCR (regular, step-up, touch-down, based mutagenesis, genotyping (Arabidopsis T-DNA mutants), 
      - RT-PCR,
      - isolation of soluble and membrane proteins,
      - solubilization of membrane proteins,
      - heterologous expression of recombinant proteins (in E. coli, S. cerevisiae, Arabidopsis thaliana, Nicotiana sp., BY-2 cell culture),
      - purification of recombinant proteins (e.g. His-, GST-, MBP-tag),
      - co-immunoprecipitation of proteins and nucleic acids,
      - purification and validation of antibodies, 
      - isolation of microsomal fraction (yeast, plants), plasma membrane (yeast, plants), mitochondria, chloroplasts, plant cell wall,
      - protein assays (BCA, Bradford, Lowry, RC DC Biorad),
      - SDS-PAGE,
      - Tris Tricine PAGE,
      - protein gel staining (Coomassie, colloidal Coomassie Blue, silver staining), 
      - Western blotting,
      - Yeast-two-hybrid,
      - VIGS virus-induced gene silencing
      - crossing Arabidopsis plants 
      - preparation samples for mass spectrometry and transmission electron microscopy,
      - circular dichroism,
