I was mainly involved in the search for new vaccine candidates against Mycobacterium tuberculosis by the characterization of M. bovis BCG transposon insertion mutants of virulence factors and in the in vivo screening of the therapeutic value of IL22 glycoforms in a mouse model for alcoholic liver disease.

Department for Molecular Biomedical research - Mouse Genetics in Inflammation Unit Tumor necrosis factor (TNF) is a powerful pro-inflammatory cytokine that plays a key role in several inflammatory diseases, such as sepsis and autoimmune diseases. TNFR1, the main receptor for TNF, is involved in inducing and maintaining inflammation, cell proliferation, cell survival and apoptosis, while TNFR2 functions in the development of regulatory immune cells and initiates healing after inflammation. Despite the successful use of TNF inhibitors to treat patients suffering from these diseases, a significant proportion of the patients do not respond to anti-TNF agents and long-term use of these drugs can cause side-effects, such as opportunistic infections and the development of additional autoimmune diseases or lymphomas. More specific targeting of the pathological actions of TNF could be possible by specifically blocking TNFR1 signaling while leaving TNFR2 signaling intact. The primary objectives of my doctoral research were to study the molecular and genetic regulation of TNFR1 and to generate nanobodies as specific TNFR1 inhibitory tools.
