    • Worked with the topic entitled "Genetic evaluation of Infertile males by using STS-PCR technique". 123 semen samples were collected from infertile males, out of 123 some were Azoospermia, Oligospermia, Normospermia and Asthenozoospermia. Morphological analysis of sperm has been done. It showed number of Head, tail , neck defects like detached head, knobbed head, zigzag tail, bent tail, proximal cytoplasmic droplet.
    • The experiment included isolation of DNA from collected semen samples using Organic extraction, Precipitation of nucleic acid, Washing to remove interfere substances, Dissolution in TE Buffer. To check quality of DNA agarose gel electrophoresis has been done. To check quantity of DNA Nano drop technique has been used which showed graphical presentation with quantity of DNA in ng/uL.
    • For genetic evaluation of DNA was subjected to PCR amplification using AZFb (sY127, sY134, RBMY) And AZFc (sY255) specific STS primers. Reaction components and cycling parameters were optimized in such a way that it remains uniform without affecting band resolution and specificity. Y chromosome specific marker SRY used as internal controlSeminographic and morphological profile of 123 infertile males revealed that there was an involvement of structural defects in spermatozoa.

     Genetic evaluation of AZFb and AZFc regions by using STS – PCR technique showed absence of deletions in both the sub regions. This was due to non genetic factors involved in male infertility.
