    The goal of the study was to determine the functional impact of a RFC1 mutation identified in a high risk prostate cancer cohort. This mutation is a lysine to arginine conversion of amino acid 627 which is located in the PCNA-binding domain. We want to determine the functional consequence of this mutation in DNA repair after UVC irradiation of cells in culture. We also tested whether this mutation influences the ability of RFC1 to interact with PCNA, and whether the mutation affects loading of the sliding clamp.
